Run your game on them, if they are taken, they will tell you and you won't have to ask. The exception is if they are planning to cheat.

If you are getting rejected in 10 seconds, then your problem is that you suck at opening. This used to be me, and it's fairly easy to fix. I like viewing it in steps. I see a cute girl, it's a nightclub, I've had a few beers, and I want to get laid. Yes, I do want something from her (in this case, to hookup). So my move would be to go up to her, make her excited to talk to me, and then follow the natural progression of steps from there to eventually sleep with her.
