Dude, college is the easiest place to get a date. Doesn't matter if she's with her friend or not. Just go up to the both of them, smile, and look directly at her and ask for her number. Call her up and tell her about a party, ask her to dinner, see if she'd like to check out the drum circle, whatever you think she could plausibly be interested in. If she brings her friend, great. Talk to the friend. It'll piss her off every second you're not paying attention to her when she knows she's the one you're interested in. She'll want you more, and it's attractive if you can have good comfortable conversation with lots of people, especially her friends.

edit: And you don't need to isolate her until it's time to get intimate. And then any of the old standby's work. Take her by the hand and say, "Hey I want to show you this cool thing." If she insists on bringing her friend, you need to keep working the situation. If you've been increasing the temperature properly she'll dump her friend in a heartbeat.
