If your interested, hit them up. Personally, I prefer not to 'play the game' with social media, I just dont like it and try not to use it. But I do keep my Instagram upto date as I know, the first thing someone does after they have met a new person, is stalk them on social media. Anyway, when I do hit them up on social media, I just start with a simple opener. I go either, Yo, do you know .....( random name of person), or even Yo, do go to (random gym, cafe, college, place). And when they say no (cos they will, thats the point of it) say something like 'oh, I would have sworn I saw you guys together, whereabouts are you from?' or 'Oh, I would have sworn I saw you there, whereabouts are you from' and then just build a convo from there. And then just chat them and get to know them, find out what there into and stuff. Keep replies consistant. And keep it light hearted and full of banter.

Just say 'Yo, I feel like I know you from somewhere'. And then if they say 'oh really, wheres that', say 'do you go to (fake place)' and when they say 'nah' say 'oh, are you on tinder?'

You should focus more on face to face, or just do Tinder if this is what you want to do.

So with instagram game, how do you hit girls up in conversations?
If by this, you mean, "How do you open girls you have never met, over IG?" There are issues with this.
You're dealing with people who you haven't met and haven't established baselines for their personalities or how they might interpret communications.
You can't see their faces or reactions in real time, the way you could if you were interacting in person. This limits your very necessary ability to calibrate and make adjustments in the moment.
Misunderstandings can occur and there's little way to recover often. Because if you're just some rando guy, they have no real investment in you as a person yet; no reason for them to invest in a 2nd chance if there's a screw-up in communication.
You're better off dealing with people in real life.

In specific cases, where they're from tinder, where they either follow back and/or like your pictures, and watch your instagram stories?
Doesn't really matter.

Knowing that they're likely single or down for a chat, what should you say?
You have some back and forth about something you've observed on their profile or pictures. If that goes well, you ask them out one on one. Drinks or coffee are easy things. Other than this, there is no real ONE thing you "say."
__4: IG is great and all, but not necessary, also__
I will say that a lot of girls will look at your instagram account for social proof that you are a cool guy.....If you develop your personal brand a bit....
Look, I don't poo-poo people who really want to post their lives for anyone to see on the Internet. You do you. And if you want to post personal stuff on the Internet, I get it. It's fun. And it can apparently not hurt to have something out there that shows what you're about. I can see the benefit of it.
But as someone who has not bothered with an IG account at all, I make for an interesting control. Not having IG hasn't affected me socially. I don't spend any energy on it, energy I have for real life social interactions. It hasn't prevented me from having a social life; I still date regularly and maintain at least 3-4 social circles.
The only really required app right now, is Facebook and a rotating cast of the texting apps, depending on where in the world someone is from. (Whatsapp, Line, WeChat, etc.)
__5: Lack of IG, doesn't hurt__
Don't get me wrong, IG has a pretty healthy adoption rate in the people around me, but not once has not having it hurt me. I think I've been asked a few times at most, if I had it. Conversation just moved right along after I've said no. Hasn't affected anything, in terms of "game."
If anything, I'm not pigeonholed by potential misconceptions about stuff I've posted, because I haven't posted anything more recently than 3-4 years ago. Except for when other people tag me in photos, which I try to avoid, especially when I'm out at clubs or bars. The result is people can only go off of the information I provide in person, which I can see the reaction of, in real-time.
People also have to spend a little energy to get information about me because it's not easily available. Like they have to interact with me directly or other people who know me. Both of which gets back to me.
__6: Less complicated__
If you intend on dating more than one person casually, it's also less complicated to NOT have a record of places you go.
__7: Internet doesn't forget__
If you want to develop a personal "brand" and spend energy working on your image, you can do that. But it's really not necessary, also. Just take care because the Internet never forgets. I think back on how a lot of older people end up with tattoos they later regret.
How many people on IG are going to post something that comes back to haunt them later? Just take care, people.
