I have never had to ask this question.

If they are in a relationship and they want me to know, they'll tell me. Even if they don't tell me, I can usually tell without them having to say anything: their boundaries will be evident.

If they aren't in a relationship, then even asking is pointless.

Open her, then ask:

1) Do you have a boyfriend?

2) Do you think I'm attractive?

3) What would your excuse not to kiss me be?

I'm just fucking kidding... Only go in for the kiss if you're extremely confident in yourself. I haven't even tried this yet. I usually ask the first two questions and follow up with, "Why the hell didn't you talk to me then?" (Say this with a smile and a really good sense of humor to not come off as an asshole.)
