Aside from buying him a drink, befriending him is a common way of defusing alpha males or beta protectors. Her brother falls into the second category. If you act like the level headed male, both of them are pretty much forced to respect you.

You don't have to buy anyone drinks but you're completely ignoring the fact that when someone is not alone, their behavior will be influenced by their status in that social circle. Friends/relatives can and WILL cockblock you more often if you don't introduce yourself.
By meeting their social circle you build up the social acceptance for her to hook up with you.

What is your first reaction to hearing she brought her brother? Personally, I'd laugh and make fun of her for cockblocking herself.
I'd ask what if Justin Timberlake (or whoever) was in the club, she was going to have a shit time because there is no way she's going home with him with her brother there.
So what you are doing is presenting a frame that her brother is going to stop anything from happening, and then she'll fight against that frame and being like "No he is cool, I can make out with whoever I want, and I can go home with someone".
After this, you have implicit permission that you can escalate without issues, because she's agreed that it wouldn't be a hinderance. That, and bring the brother into the conversation, treat him with respect. Tell him he is a good brother that he is cool with his sister having fun, etc.

I've been in this situation a number of times. Being the big brother out with my sister(s). Honestly, I'd be annoyed if you made out with her, right in front of me.
If you walk up to me, shake my hand, and shoot the shit for a minutes. Then it's fair game to chat her up all you want. But making out right in front of me, or groping her, or something. Hell no.
