I'll give some general advise and see how you respond. We can take it from there.
A couple points first:
With that out of the way...
As a rule of thumb, if you are interested in someone, then you should flirt. That's how you indicate you're interested in someone... If you don't know how to flirt, that's a different issue -- there are plenty of potential resources for this that you can look up. It's a skill that needs to be practiced.
So, flirt with her and see how she responds. If she reciprocates, that's a good sign. If she doesn't reciprocate / acts negatively, then maybe let it go (although some people are very obvious to when they're being flirted with). Assuming all goes well, you can slowly escalate things (breaking the physical touch barrier, for example).
Eventually, if sex is your goal, invite her to watch a movie with you or something like that and try and make a move.
Keep in mind: rejection is always a likely outcome. You can't fear rejection if you're a straight male seeking casual sex...
Also keep in mind: if you know her well / she's your friend, then suddenly starting to flirt can be jarring. See the point (2) above.
