In other words:

You are going out to practice. You need to decide ahead of time what you are going to say. The goal is not exactly to pick her up, but simply to get more experience with your anxiety.

If you're charming, witty and funny as you say, you should be able to come up with some interesting opener. It is not really important what it is, but it's obviously better if it spins her gears. You could say something like: "Hey, I just saw you, and this is crazy, but I think you're super cute, and I just had to talk to you. So, what are you up to?".

That is all you need to do. Her response does not matter. Your mission is simply to approach and start a conversation. Maybe she isn't interested. Maybe she's in a hurry. Maybe she thinks you're ugly. Maybe she thinks you're super hot and wants to fuck you right then and there.

In any case, you will find that not all the girls will run away screaming. However improbable this may sound, some girls will actually seem interested in talking to you. What a concept!

Also, what are some ways I can tell that my charm is beginning to win her over?

She does not run away.
She answers your questions.
She initiates conversation topics (besides "HELP! RAPIST!")

There is also a whole heaping of body language stuff you can apply, but honestly it is not important. You are a man, you think she is hot (enough, at least), and you want to figure out if she is interesting enough that you want her number. If she is, you ask for the number.

If she says no

That just means "I'm not impressed enough yet" or "Let's talk a bit more". If she was honestly not interested, she would not be wasting time on you.

Your case isn't exclusive but you lack a lot of detail for a lot of issues. Instead of giving you ambiguous unhelpful advice:

Are you clean? Cleanliness = ability to look after others = protector. People think unclean (oily hair, unkempt beards, dirt, greasy, smelly) is creepy, it's biological.
When are you having these interactions and what are you talking about?
When was the last close of any kind, and what did you close?
You're making a lot of assumptions about your charm. Perhaps it's the reason/way you're going about it, you lack detail, I lack a response.
