What's wrong with obvious? You're obviously interested in her aren't you? I had the same situation back when I was taking O-chem during undergrad. I went and sat next to this hot brunette (the only hot girl in the lecture hall, because science) and started talking. We ended up studying together at her place... let's just say we had a lot of chemistry together and there were a lot of "nucleophilic backside attacks" with cum being the usual "leaving group."

Well, lets say I sit a row behind her, how should I start a conversation with her? She's often with a couple of friends, while I'm with a friend.

I would sit on her row or the row in front so she can see me, but if you sit behind her then answer lots of questions when the prof asks them to the class and ask good questions while you're at it. It draws attention towards you and she'll likely turn around to see you talking. I don't recommend doing this unless you know your shit and you're acing the class, otherwise you'll just come off as one of those annoying idiots who ask a lot of dumb questions and give wrong answers. Also, you can use your friend as a wingman and go mingle with her group right before lecture starts or right after it ends.

Does she always sit in the same spot? If so, sit right next to the seat she normally takes. Then you can ask her if she got a note you missed or some other general question. I would tread lightly, as guys hitting on you in class can be creepy and just not the time/place. Plus, everyone can hear your entire conversation so she might not feel comfortable flirting even if she wanted to.
Since you can't walk and talk with her after class because she stays in the room, I would become class friends with this girl. Then get her number for school stuff- see if she wants to work on the online homework together, etc. I wouldn't try asking her out until outside of the classroom.
Well, as the lecture hall has lots of empty seat, I reckon she would just select another seat if I'm sitting there

I would sit in FRONT of her, so I'm in her periphery. Then make NO eye contact during the lecture, but participate (stand out?) in the lecture. Try to own the room. Then, when I'm leaving, catch her looking at me, make strong eye contact, say hi & introduce myself, and leave ASAP. Then try and find her outside of the lecture, around campus.
