Great place to meet intelligent broads who're often browsing just for the heck of it. Unfortunately, I don't see any resource discussing it as meticulously as, say, street game has been discussed.
I've been dabbling a bit with bookstore daygame, and here's what I've come up with so far -
1) Observe the woman
Probably best not to approach when it's heavily congested around her. Observe what she's wearing, the kind of books that she gravitates towards, whether she's with somebody.
2) The Opener
Experienced veterans may call me out on this, but I feel that a hardcore direct opener (the kind that Krauser and Yad do on the streets) puts too much pressure on her. Chase Amante suggests opening with something innocuous, and going "Well, I just saw you from over there and I thought you were cute. So I came over to say hi." as soon as she replies. I haven't tried it personally.
Openers that I've used:
a) Me: "What're you reading?" Her: Oh, it's X, by blah blah blah. Me: "Really? You don't look like an X kind of girl. I would've said you read more of stuff like Y."
b) "You look bored out of your damn mind." (Used it for the first time today; came up with it on the spot, since she did look pretty fucking bored. She laughed, and was pretty receptive to the conversation. That's how the powers of observation can and will work for you, if you choose to exercise them.)
I don't have a lot more to offer at the moment. I usually follow by showing her around the bookstore a bit, recommending certain books and finding out about her taste in literature. Then, I direct the conversation towards other topics, introduce playful banter that (I think) makes my intent known. What I have been exercising comes down to spontaneous conversation skills, while vaguely using Yad's Attraction - Rapport - Seduction model.
I'm average at street daygame, the weather here is unfavorable for street sarging. However, I do frequent bookstores, and I love meeting women who read.
Experienced sedditors, do pitch in any wisdom you may have to offer on the subject.
