Can women shit test you with their body language?

I've never really had this come up before, but I think I girl is shit testing me with her body language. I didn't want to talk to the girl in my class today because she had already rejected me before, so I figured it wouldn't be worth my time, but she approached me, and said: "Hey, how was your summer break?" I: "Great," then I proceed to sit down on a desk. She kept starting at my eyes, so I just asked her about her summer and then ended the conversation. Later, she overheard me ask another girl for her number to go on a date, and the girl literally turned around and stared me down in my eyes for a while.

I'm almost 100% the girl is not interested in me (because if she were she would have asked me out or not turn me down twice), but I'm curious to hear some good old RP on whether that was a body language shit test, and if so, how do I respond.
