Women are intuition machines. What I mean is, they've got a ton of processing power in their subconscious which analyzes, decodes, and informs them (at a subconscious level - they're not necessarily consciously aware of it) as to why you're saying/doing what you are. And like it or not, our motivation "leaks out" in lot's of ways we're not aware of.

Because of this, the most important aspect of a compliment is why you give it.

Perhaps the only reason to complement a woman, which won't turn her off on some level, is to express your feelings of appreciation of her. Any manipulative intent (to hold her attention, to generate a positive view of you, to make yourself less nervous by breaking the ice, etc.) will tend to reduce her attraction to you. While a genuine expression of feeling will tend to increase attraction (because it lacks self-consciousness and insecurity).

So to answer your question, the best time to give a compliment is when you feel it.

Dude. Compliment everyone freely. No matter if it's your girlfriend, your mother, a hobo, a fat chick, a cashier, a hot girl, a buddy, whatever. Compliment everyone.

Make it a habit to notice things you enjoy about people and give them compliments purely for sake of making their day better. That'll translate to perfect and genuine compliments to your girlfriend.

When complimenting things women already know they're doing well (e.g. appearance on a beautiful woman) stick to indirect or teasing compliments. For things women wish they were good at but have doubts (e.g. intellect on a beautiful woman), direct compliments can be more powerful.

Indirect:

"Not many people get this, but I suspect you'll know what I mean..." (This is surprisingly powerful)
"Would you rather be ugly or poor?" (A silly hypothetical, but by inference, they aren't ugly)

Teasing:

"I've actually been impressed with your knowledge so far, but here's the big test to prove how smart you are..." (the 'actually' is a minor neg, this compliment encourages playful banter)
"If there was a beauty contest, but only for legs, you'd win." (again, playful - hopefully she'll ask why the rest of her body wasn't included)

Direct:

"That's brave, not many people would admit that. I like that you're not scared to say exactly how you feel." (This is nice because women are rarely complimented on something like courage. It also invites them to talk about themselves).
"Am I right in thinking you were one of those quieter girls, who didn't discover how pretty she was until after she left school?" (Since it's a question it doesn't put her on the spot so much - but yes, you're calling her pretty)

Avoid stock phrases that kill the thread of a conversation; at least until you're already intimate. For example "you have beautiful eyes."

When you say something positive about her because you feel it's true.

Most men will give compliments because they feel they can get something in return and in turn, rarely get anything back. They shouldn't be used as a way to win anything from a woman because it won't.

A general rule of thumb is that compliments are acceptable as a way of rewarding somebody for their behavior.

Review:

Girl says you don't love her. That's bullshit. Don't compliment her and let her believe that she just has to put down your relationship to get what she wants.

Girl says she's glad that you love her. Acceptable. This is a healthy thing for her to say and complimenting her allows her to know that healthy behavior is the only way to get positive attention.
