No, talking to cashiers or any sort of woman who is paid to be nice to you does not count as practice picking up girls.

However, if you are practicing your general conversational skills with people then yeah that is fair, and it does help.

Sure. Your brain doesn't know that these people are paid to be there so people who are good practice. Just always remember that they are there to work so don't expect to be able to push it further then conversations much often. Practice getting into playful conversations to break the state of worker-state with some playful banter.
