Does it normally diminish a girl's interest in you, increase it, or have no effect?

There is no rule for this. It depends on the eye of the beholder.

For example a female coworker recently added me on Facebook.

Could mean something. Or nothing. You haven't supplied any relevant information about your interaction with her preceding this.

I haven't accepted it because I start to think about the types of things girls do at work (gossiping, evaluating, etc).

Dude. Sitting on a FB request from someone you actually know in real life, in this day and age, is kind of like you saying nothing and walking off if someone asks you for your number. (Hint: FB messenger is pretty much a texting app at this point and you can even make phone calls through it so it's almost like asking for your phone number.)

Also, I feel that it takes away from the mystery and intrigue of getting to know me as a person.

Mystery and intrigue is kind of worthless. The very point of them is that they prevent someone getting to know you and prevents connection, which is necessary whether you want a fling or something serious. Which leads to:

Healthy people nexting you and moving on to someone who actually opens up and doesn't play bullshit games.
Unhealthy people sticking around because they are emotionally dysfunctional and willing to fill in the gaps of what they don't know about you, with their imagination. Which is fine in some ways if you only want something quick and disposable. But this tactic is more trouble than it's worth and often leaves a lot of drama in the aftermath.

Am I over-thinking this?

I use facebook only for logistics and catching up with friends. I have a really simple profile, few pictures with friends while drinking/surfing and a profile picture. Facebook is not a viable way to get a girl. If someone actually adds me I end up accepting the friend request out of common courtesy (Only if I know her/him IRL).

Interest wise, I think if you are posting a lot you risk any potentials getting burnt out on you. You can go from being a witty and clever guy to that guy who tries too hard.

If you are always online for chat, you don't create a limited time frame. Meaning girls can easily put off attempting to contact you, or assume you're not interested since you haven't contacted them.

If a girl adds you, just simply accept at the end of the day. Most people have their social networking right in their pocket. Not really any excuse other than job/hobbies to delay adding somebody. This doesn't mean add right away, but I wouldn't wait over a few days to add. If a girl adds you, typically she is interested and wants to learn more about you. Sometimes though, they can just be nosey bitches. Either way, this is a new set of eyes on you.

If you want to create mystery, lock your profile down. Make them HAVE to add you to learn about you. Make them have to add you to view your pictures. Don't be an open book.

For the most part though, you are over-thinking this. Basically your profile on a social network is a representation of who you are. Use it to showcase what is unique about you (DHV). Digital peacocking.
