Yes. You should definitely open these girls. There's tons of things you can say.

Direct - "Hey, I saw you from over there and you are so cute that i had to come and meet you" (often gets the best results)

Observational - Make a simple statement about the environment or something she is wearing. I've started conversations before just by saying "man, it is really busy in here today" or "badass jacket'. If you can say something flirty to her based on what she is doing/wearing, even better.

Blunt question - You can even just say "What are you reading?" or "How's it going today?"

Opinion - ask her for help. "Can you recommend a good cookbook?"

Deep Thoughts - I got this one from a coach and it works in almost any situation where the girl is on her own. Just say "you look like you're thinking deep, deep thoughts right now".

If you have a genuine reason to want to talk to her that isn't just "she's hot", i don't see a problem.

She's probably reading a book because she's bored. Try to make her day a bit more interesting. Just don't try to force the interaction if she wants to be left alone.

Why are you framing it as "minding her business"? Seems like a bad frame for someone who wants to approach her. Don't frame things like you are interrupting them. Women would love to meet a great guy...if you are that guy, to them it wasn't an interruption.

As far as how to approach, read the sidebar help on "approaching." Or try /r/askseddit since it si a very basic question.

I just spent my Friday night at a bookstore... With my nerdy little brother in tow... I did just fine.

Openers used:

"Hey" accompanied by eye contact and a questioning smirk until she responds. Had one girl react with annoyance but I maintained myself and she broke. "How's your day going?" Smirk, obvious masculine attraction vibe. More inane discussion. She was there with a her grandmother and ended up turning so red and being so awkward I had to put her out if her misery and let up.
My other opener? "So what are you looking for?" Chatted with Asian anime girl (probably a 6.5) about attack on Titan. English was her second language, but we were actually vibing pretty well. She was almost immediately drawn into my space, responded pleasantly to a couple very light touches- was pleasantly surprised by her slightly hotter friend. #'s exchanged yeah yeah

Why do you go to the bookstore? Just to pick up chicks? Sounds like the kind of bullshit people can smell.
