I have done it before, and there's a few things I can point out:

Be fast. You don't know if she's going to get off in 5. Then again, don't rush if you don't have to( you can even ask where she's getting off ), but remember that the clock is ticking. This is the most important thing you have to know.
You are obviously going for a number close. This girl has plans and it's on her way to meet them, she doesn't have time for you now. Of course, after you get her number, keep talking.
Just going to point this out because someone else did, but comfort is important you are a stranger; however, I never had issues with comfort in this setting, and it's( or used to be, I'm not sure ) one of my biggest weaknesses.

As a guy who tends to approach women on public transport quite a lot, here's my two euro-cents:

It's important to understand that there is a time limit. I recommend using direct stuff - the girl doesn't have time for you. She's got shit to do, man! So do you! Understand that and act accordingly.

From my experience, one of the most important things when approaching women in public transport is to be relaxed. You're just a guy doing your thing. Talk about whatever, it doesn't matter, but make sure to let the conversation flow and use just a bit of escalation.

I'm guessing seddit is primarily car-driving Americans since this isn't getting nearly as many upvotes as it should.

Based on my experience:

Simple openers "How do i get to [whatever] station?" "Sorry do you have the time?", but you need to immediately transition. Something about their outfit also works. The main purpose of the opener on public transportation is not to wow her pants off. it's to demonstrate you are a normal person who just happens to be interested in talking to her for a normal reason. If you're with a friend(social proof to show you're not a weirdo) You can get away with opinion openers which are more fun.

There are 2 main situations where you will be approaching

At the platform - This is an easier open, but the most important thing is to keep the conversation going while you enter the train. Otherwise there's kind of a natural "state break" when you enter the train.

In the subway car - If they're sitting down, the best position is to sit next to them. The next best position is if they're sitting at a corner seat, you can open them pretty easily from the side.

Earphones Girls on the subway will often have earphones on. Don't let this bother you -- just open them by speaking or touching them lightly and motion to take their headphones off.

Don't freak the girl out Girls on the subway will often "be in their own world" reading a book, listening to music or playing a video game. It's very important to make sure the girl is aware of your presence by standing in their visual range for a second or two before you open them. last thing you want to do is freak them out.

Time constraints You need to get her number before her station. You can usually tell when they're station is close since they'll start looking out the window at the station signs, start grabbing their bags, etc.

I don't have much experience with this (yet. about to move to SF) but it's all about natural openers. Being direct might not make the girl feel comfortable because there isn't really anywhere for her to go so using opinion/indirect openers probably makes sense. I'm of direct at night and indirect during the day. That's just my thought process though (and that last part is exactly the opposite of most theory), so try whatever and see what works for you.
