If she's open to hookups, you just have to be there, flirt, gradually get physically closer (if she moves away, respect it) up to the point where a kiss seems reasonable. Grab small opportunities for brief touch if it's going well - like touching her hair/sleeve/asking to see her watch. Notice if she's doing it back. If she is, just keep going and it will be an easy move. Just don't rush it - if you're not sure she's into you, she's probably not! I've noticed confident guys usually make a move around the time the 2nd drink is over. I don't know if it's a consent/intoxication thing, or if I'm just really obvious XD

One of the main ways to get girls to hookup is to ask about logistics “what are you doing later on” “who are you here with” “are you working tomorrow” etc if shes not doing much later on, isnt working tomorrow and is with the girl that is hooking up with her boyfriend in the corner... your logistics are awesome. Shes bored with nothing to do. If however shes planning on staying out late or going to another club, or has to work early in the morning or has a female friend that she would have to leave alone to go with you... the logistics arent good so you should just get her number for a later time. Also if shes slightly tipsy but not at all drunk is a good look. Sober girls will be much less willing to come back to yours or invite you to theirs, and drunk girls are just bad news. As far as being interesting in the first few minutes... remember this phrase “take a risk” you gotta risk saying some cheeky provocative things. If shes down she will be happy you are saying them. If not, then not much you were going to do would work anyways. Something flagrantly cheeky might be something like (said with huge smile) “your certainly cute as hell/look great in those jeans, i definitely wouldnt mind taking you home” once again with a cheeky grin, not some serious ass creepy look. Just basically have fun with the girls, ask logistics questions and try your best to introduce some cheeky humor into the convo. If you can accomplish all that all you have to do is come up with some plausibly deniable excuse to get her back to yours or hers that isnt for sex... the sex should still happen if you get her back to yours and make her feel comfortable.
