There's been a few times in the past where my brother's wife was inappropriate or mean to my wife. I spoke to her directly calmly calling her out on it.

My brother blew up at me angrily saying I should have talked to him about it first. He's older and much larger than me, physically an alpha but not mentally. He threw out men should talk to the men and women should talk to women. I told him I disagreed and if he can't handle his wife, then I was going to talk to her directly. The conclusion was our wives don't talk now which I prefer. It came off more as a white knight defending the "honor" of his woman.

In your situation, I personally would just have a conversation with your brother about it calmly but assertively saying your point of view. Not because your wife told you, but initiating it on your own. That would be the most alpha way to handle it.
