Don't refuse. Take the bag and put the strap around your shoulder and pose as if you wear it better with your outfit. You set others opinion of you holding the bag and if you are unflappable then that makes you sexy.

However, if you can't see yourself taking this route then when she asks just look into her eyes and say "excuse me?" with a look of genuine confusion as if she started speaking french. Then if she asks again don't speak, just change the look on your face from confusion to "Are you kidding me right now?/Did a 3rd eye just pop out of your forehead?"

Remeber, it's not so much what you say, it's how you communicate non-verbally.

I kind of disagree with the idea that shit tests are everywhere. Or rather, that they are this adversarial thing that you have to have the right answer for all the time. Many "shit tests" are just two people finding out that they aren't compatible in some way. You shouldn't be trying to have the "right answer" all the time, so much as you ought to be trying to give your answer.

The key thing that should be worked on, is not so much figuring out the right answers, but rather one's confidence.

How do you refuse without sounding like an asshole?

I hesitate to use the phrase, "shit test," but the way you respond to requests like this, DOES make an impression.

If you say, "No," it could come off like a dick move.
If you say, "No," and give good reasons, that might be justified.
If you say, "Sure," then you're possibly being too compliant. Especially if you always fall into such a role without ever drawing boundaries for yourself. (Read: You risk becoming or turning into, a doormat.)
If you comply with a playful condition (request), that can be a fun way to be nice while still enforcing yourself.
