"I have a crush on you" or "you're really pretty" should generally be saved for when you and she are alone together. "You're cool, we should hang out sometime", as well as casual physical touch, can happen after a few minutes of conversation. Nonverbal flirting with eyes and smiling and stuff can be done from the beginning of the interaction.

Are you looking for sex or a relationship? If just sex, then the sooner, the better. There's no point in beating around the bush. You'll waste less time if she's just not interested.

If you want something more serious, I think it's good to get to know her a little bit, see what makes her tick. Give her a chance to get to know you. I still wouldn't wait very long. Say you guys have ended up at a few of the same parties, you've chatted a bit, she seems to like you... Ask her out then. If you wait too long (months), she might think you're not interested or she'll start to think of you as just a friend.

You have to frame the encounter quickly but anything too explicit coming too soon will come across as creepy; saying flat out that you are looking for sex or a relationship is a turn off.

Not very long. At first meeting. By the second meeting, no later. If the woman is showing him IOIs, he needs to act right then and there.

When a woman is interested in a man, she opens a narrow window for a short amount of time. During that time, she makes it as clear as she can that she's interested in a particular man. That short amount of time can be anywhere from a few minutes to a few weeks. Usually it's a week or 10 days. If he doesn't respond, or responds poorly, while the window is open, she concludes he's not interested or is unattractive, and she closes the window. Once that window closes, it will not reopen except in very rare circumstances. So while the window is open, he needs to step through it decisively.
