If you're wondering how to deal with this situation when faced with it yourself I would not suggest op's approach.

She's doing it to test her level of control over people. Women exchange favours as means of testing status. The person who can get the most favours while giving the least in return wins and is the queen bee. Watch women and you'll see they constantly exchange little meaningless favours (Annie ask Betty "can you pass me that" followed within a minute or two with Betty asking Annie for a small favour in return)

The exchange of favours does not apply to men. That's different. Women do favours for Alpha's, and Beta's do favours for women. Period.

The first thing any women does in a social group is try to establish herself as higher status than all the beta males. Showing the beta's doing favours for her is the most common way.

They always need you to respond to the request immediately. Because its a test of control over you.

All of this applies to female co-workers (and some types of simps). Not to your female boss.

Acknowledge the request calmly, but don't immeditale say you'll do it. Don't break eye contact from your work with words like ("Hang on a sec"). Basically make her wait a few seconds for you to respond
Check if the request is reasonable (ie aligns with what you do in your job, you're the best person to fulfil it etc)

If its an unreasonable request

Politely and very briefly say no without explaination ("Sorry, I can't do that").
Immediately direct your attention away from her and back to the task you were doing. She's only doing it for the attention and you "mini ghosting" her like this completely destroys any point in talking to you. If people around her become aware she got ignored by you she'll lose status so she'll likely take off immediately.
If she tries to continue to engage you continue to say to politely no, but don't give her the reasons. Simply say "I have to do this" and point to your own work. Make it clear your own work is a higher priority than what she has requested, and continue to ignore any other point she brings up, just repeat you have to do this. Don't justify your current task. She's not your boss.
If she escalates to management, then give them the reasons you think her request was unreasonable.

If the request is reasonable

Never respond to the request immediately. Always give at least a small delay, even if responding immeditaely would make sense. Make her wait. The longer the better. Always make her wait. ("I can do that this afternoon", "I'll be free in fifteen minutes")
She's trying to show everyone she has control over you. But the one who waits is the one without control, so she'll hate being in this spot.
Most of the time she'll try and find someone else to do it for her because waiting makes her feel lower status than you which is the opposite of what she wants to feel. She'll eventually stop asking you for stuff because it doesn't work.
As you fulfil the request make a stupid meaningless request of her while you're doing it. So if you're carrying a box for her, ask her to bring your coffee or pen along for you or something just a meaningless. This puts her in a dilemma. She was trying to win some status by getting a favour out of you, but that gets wiped out if she has to do something for you too. Make sure you do it at the same time, and use the wait time to plan what it will be.
She'll either do what you ask and feel she got nothing out the whole situation since the net favours balance is still equal, or there's a chance she'll try to bitch queen it out and just say no or fuck it up.
If she tries to bitch queen it, then call her out on it. Politely but firmly. ("I'm asking you to do something very simple while I help you. Is there a reason you can't do this? It seems very impolite to not help when you're the one asking a favour").
Bitch queen approach depends on not being called out. If she's able to act like a bitch and no-one says anything, she wins status points. She's so pretty and imporant people let her get away with shit. But if she gets called out, she loses status. So you have to call her out (but without getting angry or upset)

If you do all the above then women will generally stop asking your for meaningless favours. Female co-workers do this shit constantly and its just someting you have to deal with in a modern workplace. You can try to take op's approach but that's stupid. It will give you a justice boner but that's about it. The women won't leave you alone. They'll covertly group together and undermine you since you've made it about gender and are hence attacking all of them. That's not going your own way. That's attracting all the female attention and making your life harder. Do what I suggested and they'll leave you alone which should be what you really want.

You may feel all this stupid shit is a waste of your time and you shouldn't have to deal with it. Mosquitoes are a waste of your time too. They have noting to offer you and you just want them to leave you alone. You still have to learn how to keep them away. They won't go away just because you don't want them around.

If it's small, then write an email message to the manager saying "Hi Manager, To follow up on our conversation, there are two issues. The first is that I feel like people in the office expect me to do things because I am a man. I don't mind helping but OB was really aggressive with me and kept interrupting my work. I don't think my work should be continually interrupted just because I am male. The second issue is that OB's expectations turn into hostility when I don't do whatever she wants. She is not my supervisor and should not be telling me what to do, but she does and then gets hostile. This creates a hostile environment that I am uncomfortable with. Is it really that big of a deal? I'm a tradesman that works with women occasionally in what is traditionally man's work. Every now and again I get bitched at for being 6'3" and built because a less physically capable co-worker is doing the harder part of a task (usually by their own choice) and even I think using genders as an excuse is a bitch move. I firmly believe that if you're going to be an electrician you better be able to do the job but even that excuse won't get very far. Sometimes what happens when you grow up and be an adult is you have to do things you don't really want to do or say things you don't really want to say to maintain steady income and not be a troglodytic social pariah.

I have no idea what you do for a living but I can almost guarantee that whatever it is can't be that hard or fast paced to the point where you can't just throw some hands on a box. It's a golden opportunity to make this woman work too because you tell her you want her help since it's her box.

This is the hill you want to die on? Not helping a coworker because your men going the way of the dinosaur sensibilities might be offended by doing it? That's an 8 year olds mindset.
