A nice guy is an informal term for a teenage or adult male who is gentle, compassionate, sensitive and vulnerable.

What is the biggest factor that contributes to a guy developing into a nice guy, instead of growing up to be a leader minded guy with alpha behaviors?

Personally, I think more than genetics it has to do with an abundance of sugar coating while growing up. When i say sugar coating, I mean that the parents are too quick to dismiss unsatisfactory results in activities a boy does, such as poorly performing in school, sports or clubs. There isn't enough emphasis on the implications of being lazy or apathetic about hard work, and the parents often instill a habit of too much hope in a boy, such as "Don't worry sweety, I know one day you will do something amazing in the world." Vs "Sweety, you are never going to be anyone important or successful unless you put in the work for it." In addition, I think parents who instantly gratify boys with requests they have for gifts perpetuates a lack of confidence and determination to achieve things.

I think the above patterns lead to nice guy trends like not approaching a girl they find attractive and hoping the girl will make the first move when in fact they probably wont.
