How old are you? Because if you're a teenager, odds are a large percentage of your friends are in the same boat as you. Try to take it as lighthearted ribbing rather than some kind of malicious test, maybe even make a joke out of it yourself. Don't be butthurt about it; that just leads to more ridicule. If it were me, I'd shoot for something like this:

"So, are you a virgin?"

"What do you think?"

"I think you are."

"Deflowered a lot of men in your day, have you?"

"What?"

"You seem to have an eye for spotting an intact v-card. So I'm guessing that means you've got a bit of experience under your belt."

That shifts the focus off of you and puts her on the spot instead. Most girls are very self-conscious about looking slutty, so if she thinks she's being perceived as whorish, she'll back the fuck off real fast.

The girl that I lost my virginity to held the fact that I was a virgin over my head the entire time that we dated. Worse still, a friend of hers was interested in me (but I wasn't in her), and didn't want to hear about us having sex. BUT, she loved hearing stories about her sexual escapades.

So, yeah, I had a girlfriend running all over the place talking about all of the other guys that she'd hooked up with.

I dated that girl for a little over a year and ended it with her.

Do yourself a favor. Don't get involved with girls who behave like that towards you. If she doesn't respect you, then forget it. Don't even hookup with her and move on. That business will erode your self-esteem.

This is one of those questions you don't give a straight answer to until you're naked together in bed and her buying temperature is so high nothing is going to derail things.

One way to divert is to admit "Technically, yes, I am still a virgin." This will then make her wonder why it's technical, which you can then just say "Wouldn't you like to know?"

Like always, just keep it playful, but dodge giving a straight answer until you have established a relationship.

Just rock it and be like of course. You'll only be put down by it if you let them put you down. I went to a party where there were a couple of dudes acting hard and calling everyone virgins. If you're at the point where they're going to make fun of you no matter what you do, then it doesn't really matter what you do. Getting angry over it isn't going to help matters. I try would get them to understand that it's a personal matter and they really don't have any business asking. "So, are you a virgin?"

I think for the best effect, you'd have to deadpan the entire thing. Like you're describing a science experiment and not like you're seriously making an effort to pick her up. Maintain eye contact, don't make creepy eyes, don't look her up and down. Just imagine trying to figure out what time to get to your buddy's place. Don't do cheesy pick up lines that turn out creepy (let's play army so I can blow the hell out of you) or anything that makes it worse. Be courteous, even. You don't want it to turn into sexual harassment.

The most important part is to remember that it's a joke and play it off as a joke. Don't expect anything and don't give anything. When asked why you responded that way, you can explain the question that got this started. If she says she's joking, that's fine. You've been joking all along.

It was a ridiculous question to begin with. So I don't feel bad giving a ridiculous answer back. If you're going to brush her off and you're going to get made fun of anyways, it might as well be "I asked if xxx was a virgin and xxx thought I was volunteering."

TL; DR: "Why? You volunteering?"

If she calls your bluff... that's a different problem altogether.
